Chemiosmosis and proton gradients — especially in novel experimental contexts

H420/03 2024 Q4 (c)(ii) was identified as 'the most challenging question in the examination'. Candidates failed to reason that thylakoids pre-incubated at pH 4 would have no proton gradient when moved to a pH 4 solution (solution 2), whereas moving to a pH 7 solution (solution 3) would create a gradient enabling ATP synthesis without light. 'The majority of candidates suggested ATP production would be higher in solution 2.' This requires applying chemiosmosis principles to an unfamiliar scenario rather than recalling the standard account.

T-lymphocyte activation — receptor binding, interleukin release, clonal expansion and differentiation

H420/02 2024 Q19 (a)(ii) 'differentiated very well'. Many candidates 'did not gain the second marking point because they did not refer to receptors on the T-cells'. Use of precise verbs was essential: 'activate' and 'differentiate' are the required terms, not 'stimulate' and 'become'. Candidates confused T and B lymphocytes and described T-killer cells acting directly on pathogens rather than on infected host cells.

Homeobox genes — linking general roles to specific brain development examples

H420/03 2023 Q6 (a LoR): 'few candidates gained Level 3 overall'. Candidates who described homeobox genes as 'highly conserved' or '180 base pairs' and the lac operon mechanism filled their answer space without linking to brain development. Level 3 required specific examples such as determining the placement of the cerebellum and medulla oblongata, controlling apoptosis and mitosis to shape neuronal pathways, and switching on/off genes during neuronal differentiation.

Loop of Henle — naming the correct limb and specifying ion transport versus water movement

H420/01 2023 Q19 (c)(ii): examiners noted that 'weak responses stated that the loop of Henle decreased the water potential in the medulla but could not explain in detail how this was achieved'. Common errors included: listing wrong ions (K⁺ or Ca²⁺ instead of Na⁺ and Cl⁻); not stating which limb was being described; referring to 'solutes or salt rather than named ions'. Distinguishing the descending limb (permeable to water, not to ions) from the ascending limb (active Na⁺/Cl⁻ transport out, impermeable to water) is essential.

Synthetic biology — definition and distinction from genetic modification, PCR and electrophoresis

H420/02 2023 Q19 (e) and H420/02 2024 Q18 (b)(i): both years showed this as a gap. In 2023, 'very few candidates gave a concise description of synthetic biology'. In 2024, 'many answers discussed genetic modification, electrophoresis and PCR at length to no credit'. Synthetic biology involves designing and constructing novel biological parts, devices or systems not found in nature (e.g. designing metabolic pathways from scratch, constructing artificial gene circuits) — not the amplification or transfer of existing genes.

Statistical reasoning — choosing the correct test and interpreting significance with reference to the difference, not the results

H420/01 2023 Q15: 'choosing an appropriate statistical test for analysing data or testing for significance continues to be challenging'. A t-test (not standard deviation alone) is needed to determine whether the difference between two means is significant. H420/02 2024 Q20 (a)(ii) showed improvement in some centres — examiners noted 'fewer responses that ascribed significance to results or data rather than to a difference'. Spearman's rank correlation (not a t-test) is appropriate for testing a correlation across a range.

Meiosis and genetic variation — crossing over occurs in prophase 1, not prophase 2

H420/02 2024 Q21 (b)(ii): 'fewer than 1 in 10 did' score the mark, and 'many responses stated, or implied, that crossing over takes place in prophase 2 of meiosis and were not given'. Crossing over (chiasma formation between homologous chromosomes) occurs in prophase 1 of meiosis — this is distinct from the separation of sister chromatids in meiosis 2.

Interpreting photomicrographs of cells and tissues — distinguishing telophase from anaphase, identifying tissues correctly

H420/02 2024 Q21 (a)(i): 'anaphase was a much more common candidate response' when the image showed telophase (chromosomes clustered at poles, nuclear envelope reforming). H420/03 2023 Q1 (a)(i): 'smooth muscle was often confused with skeletal or striated muscle or just labelled as muscle, and cartilage was often labelled as collagen or elastic tissue'. Regularly practising photomicrograph identification, including tissues of the trachea, kidney, liver and gonads, is specifically recommended.
